IVF MEDIA – RESPONSIBILITIES AND SCHEDULE

 

The primary person assigned to make media will be expected to make all TALPs and other selected media and stocks no later than Monday morning on the week assigned. The seconday person will be expected to replenish all supplements that are running low by Friday of the week assigned.  Enough of each media and supplement should be available for the entire week’s IVF runs.  The person assigned for cleaning will have responsibility to ensure that the IVF culture room is clean and that inventory of supplies for IVF is adequate.

 

A logbook of culture media and cleaning is maintained. Please use it to record each time a new medium or medium supplement is made.  Logsheets can be downloaded if needed and stored in the file in the culture room.

 

 

Specific media and supplements assigned to each person are as follows:

 

Primary

OCM (oocyte collection medium)

OMM (oocyte maturation medium)

90% Percoll

HEPES-TALP

IVF-TALP

10X Sp-TL

Estradiol

Na-Pyruvate stock

MgCl2

CaCl2

Saline (if dishwasher has not made it)

Fill out checklist in the IVF Media and Cleaning Log

 

Secondary

PHE (penicillamine/hypotaurine/epinephrine)

BSS (bovine steer serum)

BSS/Heparin (bovine steer serum+heparin)

Folltropin

Heparin

Gentamicin 10 ul and 1 ml

Glutamine 1 ml and 4 ml

Hyaluronidase

Pen/strep (penicillin/streptomycin) 4 ml

Fill out checklist in the IVF Media and Cleaning Log

 

Cleaning

Mop floor with 10% bleach

Throw out large biohazard bag in outer culture room and replace

Change water bath

Wipe the surface of the hood with ethanol

Wipe the inside of the centrifuge with ethanol

Wipe the benches, refrigerator and incubator tops and other horizontal surfaces with ethanol

Wipe the refrigerator door and seal on the refrigerator door with ethanol

Clean the incubator doors with 70% ethanol

First Week of Month:  Change water in incubator pan – add piece of copper to each pan

Make sure nothing is stored in hoods and little is stored on benches

Check to make sure inventory is adequate - pipette tips, dishes, filters, reagents, etc - everything

Fill out checklist in the IVF Media and Cleaning Log

 

In addition, the person assigned to maintain incubators should do the following each week

Check CO2 and O2 levels with fyrite

Check whether temperature in each incubator is accurate
